- A C57Bl6-based alternative is the EO771 model. I have no direct experience with that, but it looks closer to what you are looking for (Kanda T et al., Cancer Res. 2009 Dec 15;69(24):9354-9).
- If you are interested in monitoring the interactions with the host immune system at different stages of tumor progression, the best model would be the FVB/N-Tg(MMTVneu)202Mul/J transgenic strain (http://jaxmice.jax.org/strain/002376.html).

Hi Olivia, I have used E0771 cells, injected s.c. in the right flank. They grew well in vendor-purchased C57BL/6 mice but do start ulcerating when they became large. Implantation number was about 1 million cells/mouse and did not require matrigel. PMID: 19118005
Ideally you inject into the mammary fat pad. Tumors grow differently in differently anatomic regions. If you are interested in breast cancer biology, then orthotopic implantation in second best to primary tumors such as the ones mentioned by Leonardo or Mahendra.
