Hi all,
I'm using DIV28 stem-cell derived Ngn2 neurons (DIV23 mouse neurons for comparison) and trying to stain for PSD-95.
Cells are fixed in 4%PFA/4% sucrose at RT for 15 min and permeabilized with 0.5% PBS-Tx100 for 5 min.
Washed with PBS, blocked in 10%NGS/0.1%PBS-Tx100
I've tried the following antibodies:
UC Davis NeuroMab clone K28/43, Synaptic Systems 124 011, and Thermo MA1-046.
Secondary is 1:500 AlexaFluor 488 and 594
In each case, the PSD-95 Ab's work great on mouse primary neurons, but crappy on human neurons. You can see the great mouse primary staining in the one image (poor human staining in this case could have been due to the very low amount of MAP2 positive neurons and therefore limited connections), but in the second image (showing only human and with more MAP2+ cells), there is a very small amount of FL above background for the NeuroMab antibody, but nothing that resembles the punctate staining similar to Synapsin (shown in the corner).
I've tried to get these Ab's to work so many times but it's just not working. Does anyone have any suggestions, maybe a different fixation process or have an Ab they know for sure works in human derived neurons? Thanks so much!
