Doing some RNA-FISH and I'd like to use Cy3-Tyramide and incorporated digoxigenein-d?TP (opinions? dUTP? dCTP? manufacturer? which nick translation kit?) to do some signal amplification.
What's ya favorite protocol and where do you get your reagents? I'm looking at the Perkin Elmer ( https://www.perkinelmer.com/lab-solutions/resources/docs/BRO_TSAsignalamplification1.pdf ) and the Enzo kit ( http://www.enzolifesciences.com/science-center/technotes/2018/october/shall-you-use-biotin-or-digoxigenin-for-ish-probe-labeling?/ )
Do you modify anything? Use home brews for buffers? Are you quenching endogenous peroxides first and doing a Proteinase K digest?
