I am wandering if anyone has a recommendation for the proper concentration of 4-hydroxytamoxifen to induce CreERT2 expression in primary pancreatic ductal organoids generated from uninduced (no tam administration) mouse pancreata?
Have successfully generated the organoids and was wandering if anyone else has a recommendation? I treated my first well of organoids with 100nM of 4-OHT (from concentrated 4-OHT in ethanol @ .05mg/mL, or ~129uM ---> 3.875uL per 5mL of organoid growth media).
100nM seems like a decent starting point based on the literature I have read, but I have seen some papers (intestinal organoids) using as high as 1uM [4OHT].
Any input or advice would be appreciated.
The more, the merrier. You want to maximize that all CRE-ERT2 cells properly act on any available floxed sites.
