I am currently working on patient derived organoids from pancreatic cancer (PDAC) patients. I noted that my organoids never grow too much. after 1 month in culture they are usually around 100-150 um. I am following this very famous protocol (Article Organoid Models of Human and Mouse Ductal Pancreatic Cancer
).Do you have any suggestions?

What do you know about the tumor itself? I assume these protocols are not an one size fits all, I would try to adapt the protocol and the mixtures on the pathology of your tumor and its transcriptomic profile. This won't be easy, indeed, establishing such an algorithm for every tumor. I have never worked with pancreatic cancer organoids, but I had the same problem with urine bladder cancer organoids. Hope I helped a little bit
Look at your growing organoids too, they tend to shift their gene expression as they grow and they might have other needs.

Hi Giulia, one of our recent collaborators successfully produced PDAC organoids approximately 800 µm in diameter. You can check out their publication here: Article In vitro Characterization of Insulin−Producing β-Cell Spheroids
