Is it possible to attain IC50 if my target cells have less than 50% antigen expression ?
For example:
Cancer cell line XXX is showing 35% of expression of cell surface antigen YYYY,
if i target this cell line with ADC that are specific to this antigen, will it give IC:50 ?

Theoretically yes, but it depends on the drug conjugate and its mechanism of action.

Absolutely yes! The cells expressing the specific antigen, will be initially damaged/killed by the antibody-conjugate. Then, according to some thing like the "bystander effect", other cells, even those don't express the surface antigen, will be affcetd by the death ligands, reactive oxygen/nitrogen species and so on, released from the damaged/dead cells. Hence, it is certainly possible to obtain IC50. Good luck.
Thanks for your kind response, i do agree that bystander effect can increase the % of cell death but not at substantial amount . i.e 20% ADC targeted cell death can give 10% bystander effect but not 30% bystander effect, purely based on my speculation.
To support this speculation i found article "Clustering of apoptotic cells via bystander killing by peroxides" -2000 REZNIKOV.
My conclusion is there is a possibility that 35% of ADC cell death might or might not give IC:50 (In vitro) via bystander effect.
