I assume you are referring to a commercial kit for HLA B27 typing using PCR-SSP method. During PCR-SSP amplification, an internal control primer pair is required to be with specific primers in the same reaction mixture in the test tube to ensure that if amplification failure occurs, it results from mismatched primers rather than from a failure of the amplification system. The internal control primer pair specifically amplify a non-polymorphic target, usually a fragment of single copy gene on both haplotype. Inno-Train’s HLA typing kits use HGH (human growth hormone gene) as the internal control, but it does not have to be HGH.
Thank you so much mam for such valuable answer. I am not using kit but designing and implementing Allele specific PCR for HLA B27 typing and upon literature review and studying previous researches. HgH WAS USED AS INTERNAL CONROL IN MOST OF THE CASES, so I asked this question. Thank you again Mam.
