According to Kokubo's instuction for preparing SBF (Simulated Body Fluid) pH should be increased from 2.0 to 7.4 after adding Tris to the solution. I added all additives one by one, and the pH became 1.8-1.9 right before adding Tris. But then after adding Tris it didn't increased.
Does it mean the Tris composition is wrong? or Should I consider something especial in adding Tris?

I have a few questions that might help clear things up:
1. Did you observe any pH change upon Tris addition?
2. Are you using Tris base (CAS: 77-86-1) or Tris-HCl (CAS: 1185-53-1)?
3. Did you observe the formation of any precipiates during Tris addition?

Where did your TRIS come from? Is it free base or hydrochloride?

CAS No is 77-86-1. (It means that it is Tris base. right?)
I didn't see any precipitation during preparation. But the pH didn't change at all.

My Tris is from CDH company which is an Indian company.
I checked the Tris like this:
Adding Tris to distilled water and in another experiment adding tris to water after adding 1.0 M HCL. In both cases pH was increased but may it has buffered after adding other components like KCL, Na2SO4 and ...
