I started to culture potato eyes in a tissue culture medium and got one growing shoot. The problem is there are very thin side shoots of this growing potato. Does anybody know how I can resolve this problem and induce the shoots to grow thicker? I use MS medium for the potatoes.I use LED Lights with a 40W energy saving white lamp as light source .Potato medium contain 2mg BAP , 0.5mg NAA and 1mg GA3 with 148mg NaH2PO4.this is first time that i culture potato and this thin shoot is my first and only successful potato culture .

The first hunch would be - cultures under sub-optimal light conditions. Either increase intensity or duration. MS medium is fine as it has adequate level on nitrogen.
Not to worry. After some transfers they should look better. And as Jason is suggesting: maybe add a low amount of hormones. Shoots from potatoes in the dark from eyes also look very thin at first.
I think MS is okay however, if you modify it by adding some plant growth hormones it will be much better. the second point is the light intensity and duration.

You don't need to add growth regulators, except perhaps 0.05mg/l BAP for potato shoot cultures. Even so, you can get scale like leaves, and this is usually caused by a build up of ethylene in culture vessels. You can get over this by allowing some gas exchange (eg by loosening the lid) or by adding silver ions to the medium, which inhibit ethylene production.
