Is it possible to induce somaclonal variation in ploidy level in micropropagation of Triploid Lilium Hybrids? I am interested in inducing somaclonal variation in ploidy level of triploid lilium hybrids to obtain diploid or tetraploid line. I know that Surflan (Oryzalin) can induce polyploidy in diploid lilies, but there is no published research on somaclonal variation in ploidy level in micropropagation of triploid lilies. I have Surflan from USA .

As you know micropropagation is depend on mitosis division and we never find genetically off-type plants (especially in bud culture). Then this technique isn't efficient for changing of chromosome set. I think protoplast fusion and somaclonal variation (via numerous subculture of calli) are more efficient.

I just obtain 2 mutated plant (somaclonal variation) after about 3-4 years of micropropagation and growth in soil . pink blush color is disappeared in mutated plants and flowers are wider than original variety .
I don't know if their chromosome number is changed or no but this is a good material for my breeding program ,
This is accidental mutation and no mutagen agents is used for this plants .

I think that flow cytometry will be expensive for me to count chromosomes or detect all mutations in genes . for now only way to detect mutations will be changes in appearance and flower color and forms .
Flow cytometry of sports of triploid Rhododendron has indicated that occasionally such sports can be diploid or tetraploid. However most sports of triploid Rhododendrons are triploids.
