I need a moderate to easy method and protocol for the selection of salt and lime (calcium carbonate) tolerant clones and seedlings of Lilium species. In vitro grown seedlings and micropropagated plantlets are ready for trial. For a Lilium species preservation program, I need to determine the salt and lime tolerance of Lilium species because our waters contain calcium carbonate and it seems that not all of the species can tolerant this hard water.
I have a collection of Lilium species and add new species every year ,for this research i will use Lilium Ledebourii and later some American Species Like lilium philadelphicum and other acide loving lilies
The first plant-organ affected by salt is root, it will inhibite the root elongation. You may caount the number of root produced and the lenght of the maginal root under in vitro condition.
Thank you very much friends , now i have enough information to start , Lilium Ledebourii will be first lily species that have to be tested and later North American species .Dear Dr Abdollahi , my trial must have an easy result , this species can survive and grow well without morphologically changes or no , if clones can't survive then i will try with seedling population and micropropagated clones to select a lime tolerant clones for my own lilium collection for future commercial projects.

I advice you to select from seedling not clones. Under micro-propagation conditions the plant already have its moisture from the condensated water resulted from evaporation, so you think that you have tolerant clones but the fact is that you don't have any stress. Do your selection in the field after acclimatization that will give you the real answer whether your clones are tolerant or not.
Thank you Amera , Some of my Lilies are rare and seeds hard to find , i culture seeds and i have seedlings in test tubes , i can propagate my seedlings and use them in my trial as you recommend , but i have not lots of space for field trial at first step , i use Plastic Wrap (household used clear plastic wrap ) instead of expensive parafilm and moisture is not high as the moisture in jars but i will consider your opinion in moisture inside of culture vessels and try to reduce moisture inside of test tubes .
