The Griess assay is not working for the standard curves. I am using 100uM-1.56uM standards. When I add the Griess reagent the high concentrations are yellow. Lower concentrations go very dark pink and crystals form at the bottom. What am I doing wrong?
Use fresh reagents prepared 10 minutes before using, and kept in dark. Also, water should be clean (especially of nitrite traces).

I never face this problem, Griess reagent works slightly acidic medium so Phosphoric acid should be 2.5-3% range, only one thing need to be kept in dark is Griess reagent, nothing else. In alkaline medium colour is often yellow.
