hi, I am currently isolating DNA from tissue. I am trying to analyse the DNA integrity by running a gel electrophoresis experiment. I'm currently using 1xTAE buffer, 0.5% agarose gel, 60 voltage and 10kb ladder as recommended. However my DNA is degrading but the RNA is completely intact. I repeated this gel protocol using DNA isolated from blood, which showed full DNA integrity.
Are you keeping sufficient EDTA present during your DNA purification? That inhibits most DNases by chelating the necessary divalent cations.
Thank you Michael J. Benedik , I had made sure I had no traces of EDTA during my extraction process. This was the first thing that came to mind too.
Did you perhaps write your answer incorrectly? You need to have EDTA present (not remove it ).
