Dear All,

Many suggests that it is better to run SDS-PAGE in lower voltage, for example 100V in mini-gel, espcially to separate close molecular weight bands, but I don't understand what is the reason behind this.The only reason I could think off is heat.

The higer voltage generates more heat. Too much heat can cause diffuse or smile bands. So what I think is that if can control cooler temperature, the higher voltage (eg. 200V in mini-gel) can be used and gel can be run faster. Am I right? or, Is there other reason to run lower voltage beside heat? , espicially to separate  close molecular weight bands?
