Hi all, I recently find the optimization of inflammasome studying conditions in THP-1 cells. THP-1 cells can be activated not only (non-PMA) LPS+ATP [Ref; invivogen] but also, after PMA-differentiated, LPS+ATP for activation of NLRP3 inflammasome.
What is the difference betweent the PMA-differentiated and un-differentiated THP-1 cell upon inflammasome study?
I agree with Anja's response and add that we have noted additional differences in activation of other inflammasomes without and after PMA treatment of THP-1 cells (see the link below, Fig. 3C).

In our hands overnight treatment with either PMA or Interferon beta resulted in upregulation of key inflammasome components resulting in a more robust inflammasome response than in untreated THP-1.
