I am working on Rapid test development (Lateral flow based). I am facing conjugate release issue after few months of strip preparation. Up to approx 30 days, conjugates release found proper but as strips ages say after 30 days, conjugates doesn't leave conjugate pad after adding sample. Need help.
You need to provide more information so that we can suggest solutions! Type of conjugate pad you are using, concentration of reagents you are using and drying methods.
Agree with the previous concerns over it not being stored in a dry environment.
this sounds either as a pre-treatment problem, materail problem or stability problem.
pre-treatment: Your buffer seems to be OK and should allow conjugate migration.
Material, you might be using a material that binds itself to the conjugate as it dries and ages, try something more hydrophobic
Stability, the conjugate could be aggregating slowly on the pad or collapsing. try different pH during conjugation and different stabilization agents like BSA or PEG and test the condition of each one after 2 or 3 months
