It will depend on what you are trying to do, but some of the early studies on endotoxins looked at the effects on cell viability (Lucas RM, Chen SY, Aleo JJ. Histochemical study of strain L fibroblasts exposed to endotoxin. The effect on cellular organelles. J Periodontol. 50(1):20-2. (1979)) and immune response (Erwin Neter, Endotoxins and the Immune Response, Current Topics in Microbiology and Immunology pp 82-124 (1969)).
Some E coli strains release a lot of endotoxins, some other don't. This makes many plasmid preps toxic to cells. Because the amount of endotoxins will vary within a broad range from one maxiprep to the other, getting rid of endotoxins is a good idea if you want to make your experiment reproducible.
If you do not use Endo toxin free approach, you may have some false data while you perform experiments. It has a high chance to give you that kind of results
