I want to study a immunosuppressive gene.

I knock it out in a cancer cell line and transplant it into B6, and compare its growth with WT tumor. I would expect that KO tumor grow slower than the WT. This is an simple experiment. 

I saw people use the same cell line but transduced OVA in it. After they transplant the OVA-cells into B6, they do OVA-immunization first, then they compare KO tumor growth and WT tumor. 

Why do they use OVA cells and OVA-immunization? The first experiment is much easier and can prove the point.
