We all know that CSC (Cancer stem cell) are germ line of tumor giving rise to body of tumor cell. Then why this body of tumor cell fails to carry single gene mutation or single gene alteration, that first  arise in pre-cancerous CSCs.

Imagine a condition where I activate one proto-oncogene (RAS) in NSC(clonal Normal stem cell) and injecting it in NOD/SCID mice to give rise to bulk tumor. Later When we do gene expression/mutational study in this bulk tumor, we won’t find RAS gene alteration in every tumor cell within this bulk tumor, Why it so ?

Does it mean bulk tumor is formed not only from clonal normal stem cell that carries single aberrant gene(RAS). (Or) Will there is any chance for the mutation (RAS) to be corrected (De novo/naturally ) after few doublings of  clonal Normal stem cell/CSC.
