Hi, We used Ficoll to isolate PBMC from mice whole blood cells, but there are many little nights,with no celler structures, in PBMC through a microscope. We guess these granules are paltelets, but we don't know how to get pure PBMC by Ficoll.
The platlets can be removed (or i should say minimize) by multiple washing after taking Buffy coat. Once PBMC (with contamination of platlets) are plated you need to wait for their natural death.
Multiple washing at low speed centrifugation after PBMC isolation is the only way out.

I am wondering whether these little round particles that bother you would be mouse red blood cells, not platelets. In that case you could get rid of them with a brief treatment with a special RBC lysing solution (I don't remember the formula but you can probably find it easily).
