i want to detect stress produced by prion strain in SH-SY5Y or N2A but i am not sure which cell line is better?can anybody help me with selection of cell line?
I never worked with N2A cell line! But I can tell you is that SH-SY5Y cells are very susceptible to both ER stress and apoptosis induction, compared to other cancer cell lines (such as hepatocellular carcinoma, melanoma and breast cancer) !!!
Classical SH-SY5H cells are a mixture of N-type (neuronal), S-type (substrate adherent) and I-type (intermediate) phenotypes !!! You can keep them and growth them as a mixute or, alternatively, you can select the N- or S-type by using or not trypsin when you split them.
I'm telling you this just because the S-type usually are more sensitive to stress.
We use N27, a rat dopaminergic cell line, for sumoylation/ubiquitin studies. They are resistant to many manipulations. You can get them from Millipore.
Is the type of system you assess relevant to the research questions you are asking? Considering that SHSY is a human line, and N2A, is rodent I think..
