I'm planning to investigate the activation status of porcine (pig) platelets as a response to various stimuli. What antibodies would you recommend for flow cytometry?
Well the best ever is not an anti-body itis fluorescent annexin V that bounds to the exposed phosphatidylserines, otherwise it could be also an anti-body against P selectine, a protein exposed to the surface only when platelets degranulate. Shall you need references for the antibodies against porcine proteins ?
References would be great! I have a CD62P-antibody, but it is present at all times and increases in intensity during activation with PMA on bovine and ovine blood. Does this sound right to you? What treatment would you use to activate pig platelets as a positive control?
Well, not shure to be able to give you the actual references of antibodies, I am not experimenting anymore (shame on me...) but you may find those in the attached paper (well, no, it does not want to be attached... see my profile, coauthors are Giraud and Beliard). No sweat with the annexin since it is not species specific... just check the adequate label withe cytoflow expetr in your lab.
CD62P may be positive, because quite often platelets are marginally acitvated during preparation AND becuase they have an Fc Receptor, then can be marginally affected by the use of wole antibodies ... use FaB'2 whenever possible...
Yes it is normal that PMA increases P selectin detection. However for full positive controls it is not enough most of the time, best woould be thrombin + calcium ionophore... but you do not want them to agregate, some kind of a vicious circle....
