No one in my lab has experience growing U937 and I would like to know
What's the best way to subculture the cells ( using RPMI-1650, 10% FBS and 1%PS), Do they attached or maintained in suspension? what do you recommend a T-flask or TCP? How many times, do I need to change the medium? How do you freeze the cells?

In brief, effectively, RPMI +10% FCS is fine. These cells are normally in suspension and should be maintained at a confluence between 10^5 and 2x10^6 cells / mL. To freeze them, add 5% DMSO to this medium.
For subcultures, Mondays and Wednesdays I leave them at 300-500k/mL and Fridays at 200k/mL. I use TPP cat. No. 90075 flasks, but any standard tissue culture T-flask should fit.
