Hello all, i am working on Chilly anthracnose disease and the pathogen involved is colletotrichum capsici but the problem is occuring while subculturing, i can isolate pathogen from chilly fruit but when the culture is grown when i need to subculture its not growing and also if its grows other organisms are growing along with please help me with this
The problem happens when the preservation method is not proper particularly long-term preservation. This is not only for Colletotrichum but other pathogenic genera such as Pestalotiopsis, Neopestalotiopsis, and some other genera. I recommend to culture the fungi on sterilized filter paper placed on the surface of PDA petridishes. You can cut the filter paper in 5 mm and leave 20 to 30 pieces of paper on the surface of the PDA. Then cultivate the fungi and after 10 to 15 days, when the papers are completely colonized by the fungus, collect the papers under the lamina and let them dry completely under the lamina under sterile conditions for a few hours with the air flow of the lamina. Then transfer the pieces of paper to the cryogenic tubes and seal well and place the tubes in the freezer. For re-cultivation, it is enough to place a piece of these saved cultures on the PDA. This method prevents the death of fungi to a large extent in the long term, and it also does not suffer from fungal bacterial contamination.
