The following (picture) unknown organism has contaminated our SH-SY5Y cell line, any idea what it is? They are only there in the flasks with cells, not in flasks with culture media alone.
Charles: your cell stock was contaminated with bacteria before. Try to wash away about 2 hrs after seeding when your cells stick to plates for a few times and add antibiotics such as ampicillin in the medium as well. Good luck.
Bacterial contaminations should cause pH shifts quicker and develop turbidity.
The waterbath would be my immediate suspect, which needs to be cleaned weekly.
Be careful about using your prepared medium, it would be wise to put media in a flask without cells and monitor any potential growth if you do not want to get rid of the media.
I am facing same problem of contamination in my cultured flask.
I am using SH-SY5Y cells (ATCC) passage 2. Media I am using is DMEM-F12 (1:1) from invitrogen, 10% FBS (Invitrogen, I SHOT FBS). 1% Antibiotic (Pen/Strep) of invitrogen.
I have also tried the cells subculturing with ATCC recommended media (EMEM-F12 (1:1) + 10%FBS+1% Pen/Strep.
Initally the cells grows well for 6-7 days but after that cells get contaminated. I have revivied the cell number of times and same contamination i am observing.
