What should be the optimal number of cells per well in a 96 well plate to perform cell growth inhibition assay or MTT assay? The cell line in question is the A-431 cell line. The cell line has a doubling time of nearly 30 hr.
The number of cells required for MTT assay depend on the period of time of reaction. If you perform reaction for 72 hrs, it is better to seed 5000 cells/well. Other wise for 24 and 48 hrs you can seed 10000 cells/well.
For your information, the capacity of 96 well is near about 1 to 1.5 lakhs/well. You can seed the cells accordingly.
Basically, for A-431 cell line, 5000--7000 cells/well in 96 well plate/48 hr is enough. However, you are highly advised to perform a cell number titration to determine the seeding density. As the number of days of your experiment will determine the number of seeded cells.
