Now many companies are providing IRD800 or IRD700 or alexa fluor 680 dye labelled secondary antibodies. What is the best? Could you recommend a company for western blotting using odyssey infrared imager?
Hi I used to use the secondary antibodies supplied by Li-cor. I have used both Fluor 680 and 700 dye for P-MAPK and actin respectively. I used to use 1:15000, 1% milk, 1 h, room temperature.
I was using Licor secondary antibodies but now I switched to Thermo pierce DyLight 680 and 800, they work well at 1:25000 dilution and are cheaper than Licor antibodies
I too have had good results with rabbit Thermo Dylight 800-4XPEG, Dylight800, and mouse Dylight680. The 4xPEG version is at least twice as bright as the non-pegylated form. I dilute 1:10000, incubate 1 hour in 3% goat serum for westerns. Waiting for 680 to be available in the 4XPEG version. LICOR Odyssey Classic is being used for imaging. By the way, the 800 must be kept wet. The 680 is actually brighter if the membrane is imaged dry, but works fine when wet too. 800 dissappears when imaged dry and can not be repaired with rewetting.
