I know that the prepared medium shoul be used within three weeks only of prepration then discarded but a college told me that the FBS is even more sensitive and should not last more than two weeks. what is your experinece?
Instead of pouring 50 mL of FBS into your 500 mL bottle of cell media, you may wanna consider to prepare smaller amounts of media containing FBS right before you need it (for instance for passaging). That's how we do it. We add 2 mL of FBS (from 15 mL frozen aliquots) to 18 mL of cell media, let it warm up and then use it. Once the FBS aliquots are thwaed, we keep them cold in a fridge. They will be used up in no time.
Another advantage is that you can add your antibiotic/antimycotic freshly to your cell media as these substances quickly degrade from frequent warming up of your cell culture media.
Honestly I've never had an issue with media that is several months old. I wouldn't use media that 6 months old, though I might if I had no other media, but generally within 3 months its fine so long as its in the fridge, not kept out, or in the water bath for too long for each feeding. Unless of course you are having major issues with TC or your cells are super sensitive. But even for ESCs I've never had an issue.
