If pH and conductivity is not the major concern then can I use 45-50 degree Celsius WFI (water for injection) for preparation of phosphate buffer. In fact, I have already known how much chemical is needed for my desired pH and conductivity. So, if I prepare buffer at 45-50 C and remain it for cooling at 25 C, will it be a proper technique?
Hi Taufiqur: The pH value may be slightly affected by the temperature change from 50C to 25C, but not a lot. You may warm your pH standards to 50C as well for calibration and may eliminate such a variation. Good luck.
Be careful not to produce a supersaturated solution that precipitates when it cools.
