What is the difference between Lipofectamine 2000 and Lipofectamine 3000 for stable transfection of Lentivirus?
I was told that Lipofectamine 3000 is more toxic to the cells with higher transfection efficiency.
I wonder which one is better for HCC cell lines.
I'm assuming you mean stable transfection of packaging plasmids to make the cells produce lentivirus? We have used both Lipofectamine 2000 and Lipofectamine 3000 for this purpose in HEK293T and there wasn't that much difference. The 3k probably had a slighly higher transfection efficiency. Can't really comment on viability because they barely last 48h after transfection regardless of what reagent is used. Hopefully there will be someone who has tried both in the specific cell line you're using since that will be more useful information :)

I think there is no obvious difference in the lentivirus packaging yield between Lipo2000 and Lipo3000. But for mesenchymal stem cell transfection, e.g., bone MSCs, I think Lipo3000 would be better.
