We have compared the global miRNA expression profiling in spleens of diseased and treated mice. What is the most popular method in terms of validating miRNA expression among the following: probe sets from Exiqon, SA Biosciences, Nanostring or Taqman probe from Ambion.
best method might be Nanostring and Taqman probe. This is highly quantitative. Nano string will be more appropiriate if quantity of RNA is very less and you can quantitate larger number of rna species in assay. Micro fludics based super arrays are also usefull in this.Further, RNA sequencing also will help in this regard but greater experteise is required in analysis.
We have performed the genomic miRNA expression profiling using SA Biosciences' RT-PCR based method. Therefore, I do not want to do another global expression profiling by using for example Nanostring. Now we want to validate selected miRNA genes. I was wondering about the experience of others who are validating the miRNA expression using Taqman and other probes. Specifically, if anyone has experience comparing Taqman and Exiqon, I would like to know.
I have had success using the the ABI Taqman assays. I haven't tried Exiqon qPCR probes, but have had good luck with their LNA probes for in situ hybridization.
