I made IHC and count number of tumor associated B and NK-lymphocytes in peritumoral zone of endometrial adenocarcinoma. Tau-Kendall analysis found direct correlations between this two kinds of cells. How can I interpret it?
That is an interesting question which is more conceptual to what is considered to be a the humoral (antibody) response to a neoplasm. I am also working in that field, having a particular project, where we identified also plasma cell accumulates in mouse neoplasms. Quite a difficult task to interpret in the context of a general dogma for cellular response ...
I will let you know if I can come up with something.
Vaccines (Basel). 2015 Dec 17;3(4):1019-51; Proc Natl Acad Sci U S A. 2015 Oct 6;112(40):12474-9. Yours is a fun question... that would take two semesters to answer correctly.
Short answer: You saw a large crowd around a looted store and you observed what looked like men and women. What does it mean?
If you had looked closer (better Ab) you would have noticed some were dressed as street-beat cops, some as senior detectives, some as looters and lawyers, and others as curious bystanders drawn there by the lights and sirens.
The tumor was able to grow because it cut off many of the burglar alarms (surface camouflage and sending out spoofs in the form of exosomes) and it called in lawyers (lymphocytes of various types to suppress tumor rejection). But the tumor makes so much 'noise' in the looting that the street beat cops (B-cells) are sniffing about, and detectives Plasma and natural killer cells) and paddy wagons (macrophages) are mingling in the crowd. But there are cops paid off by the tumor in the mix, so you don't know who to trust.
Better antibodies will let you sort out the story.
Thank you for your answer and limks, mr.Gregory S Buzard , but:
1. I used Ventana(Roche) antibodies and made all staining by protocols on Benchmark GX(Ventatna) stainer+ possitive control of staining and demask of antibodies.
2. I am intrested in correlations in this case and tumor of this localization.
3. Sorry your explain of antitumor immunity is not clear because organisation of police work in my country is not the same as yours, that is why I prefer to believe in classic theory of antitumor immune responce.
2006 slide show on tumor immunology, informative. My apologies for cultural lapse in choice of analogy. My point is that, 10 years later, in 2016, there are hundreds of excellent antibodies against truly functional subsets of immune cells that can tell you so much more about your tumor environment than just that there are B-cells and NK cells present in a coordinated fashion.... Even the macrophages have important subsets that have critical functions.
Thank you, Gregory S Buzard. Inmy research I use stain for macrophages, dendritic cells and T lymphocytestoo, but so strange correlation I got only withB and NK cells.
