I'd like to perform some experiments by applying metronomic chemotherapy with 5FU and Oxaliplatin in colorectal cancer cell lines. Has anyone used a particular concentration for this application?
metronomic means a dose which is significantly lower then the IC50 but for an extended periode of time. In calssical chemo-sensitivity assays of CRC cell lines, we measure IC50 for 5-FU in the range of 50ng/ml up to over 50µg/ml for a 144 hour assay. And for Oxaliplatin it is between 50ng/ml and 3µg/ml.
These ranges tell you that you would have to first measure your IC50 (for every cell line you use) using one defined test. And do yourself the favour to repeat it at least three times. Values depend on so many factors present in cell culture ...
Then, you can go for a metronomic long-time treatment using for example only 1/10th or 1/50th of the IC50 concentration.
