Regarding CAR-T cell therapy, during co-cultures with different effector:target ratios, when we refer to "effector", does it involve the total T cells? For example, does a ratio of 5:1 mean 5 total T-cells per cell, or 5 CAR-Ts (taking into account transduction efficiency) per cell?

In the context of CAR-T cell therapy, the term "effector" typically refers to the genetically modified CAR-T cells. The remaining unmodified T cells will not recognize the target, thus are not "effectors".

But let us take into account that you will have alloreactivity, and also, depending on your transduction efficiency, you may have to adjust the numbers. Usually with Prodigy you reach around 40% of CAR+. My question was related to how you fit your numbers based on transduction efficiency if that would be the case.
Hi Gonzalo González Domínguez. Let's say you want to do a co-cultivation assay with a effector:target ratio of 5:1. If you have 1e6 target cells, you will need 5e6 effector cells (CAR+ cells). Considering you achieve a transduction efficiency of 40% CAR+ cells, you will add to the co-cultivation assay 12.5e6 transduced cells.
