Dear all, recently i have tried to optimise my cell culture model by adding growth factors to it. The question is, which concentrations should i try? Most literature have reported using about 10 ng/ml. What would you consider to be most optimal?

On which cells are you working? What growth factor(s) is/are you using? You can try optimizing it by yourself with the help of literature by carrying out a dose-dependent response. And then if possible/feasible, then a time-dependent response with the dose that you optimize.

I agree with Debaki’s point of view. But if your question is in general than the answer is FBS or FCS. Usually, in cell culture, we use 10% FBS/FCS as an additive to your media which supports the cell growth very beautifully.

It really depends on the growth factor and cell type. If you are using 5%FBS, there is no need to add growth factors because serum is loaded with them. If you are trying to use a low serum or serum-free medium, then you need to look up a recipe for that particular cell type. For example, epithelial cells typically require the addition of transferrin, hydrocortisone, insulin and EGF at concentrations of 5µg/ml, 0.5µg/ml, 5µg/ml and 10ng/ml respectively. So you see, there is no general answer.
Unsurprisingly, the answer depends on what you are trying to do. Are you treating cells with SCF or insulin and want to maximize your response then treat with 1000ng/ml or even more. On the other hand, if you are treating a sensitive cell line with EGF yo obtain a physiological response treat with 1ng/ml or even less. With the details you provided (none) there is no way of knowing what optimal even means.
In the end, one of the previous answers captures what you need to do: figure out what optimal means for you and then run a test.
