While IC50 concentrations can inhibit 50% of cells' viability, it may have confounding effects on gene expression. Therefore IC30 or IC40 would be better choices. Please discuss about it.
IC50 value measures the potency of a drug's inhibitory effect on a biological process. It is not directly applicable to studying gene expression, though it may provide an idea about the concentration to be used.
I suggest you use concentrations covering a range both below and above the IC50 value. You could select two concentrations below the IC50 value and two concentrations above the IC50 value. This will allow you to observe how gene expression changes with increasing drug concentration.
For instance, if the IC50 value of a drug is 5μM, you may choose to use concentrations of 2.5μM (IC50/2), 0.5μM (IC50/10), 10μM (2 x IC50), and 20μM (4 x IC50).
