It seems like you can only make droplets for 8 samples at a time with Bio-Rads droplet generating cartridge. Is that true? How expensive are the cartridges you have to use to make the droplets? Do you need to use a new cartridge each time? Do you find that there are often actually multiple DNA targets per droplet, when it is advertised that there should only be one? How would you detect this? Any help is appreciated! Thanks!
The limitations depend on the subject you are working on and to which other platform you compare. Compared to qPCR for example, ddPCR has a limited dynamic range for detection (and is smaller than in qPCR), but for quantification they can be similar.
In case of AutoDG system, you can have a full 96-well plate and the robot does the whole droplet preparation process. But still, also the robot handles only 8 wells at once.
The price of the cartridges depends on the distributor. We get a price of cartridges at around 1.3€ per well. the cartridges are not re-usable.
Regarding the number of targets per droplet, this depend on the concentration of targets in your sample. Theoretically the maximum is slightly below 10 per droplet. You do not detect this directly, but calculate it based on the number of positive droplets against all droplets and by using Poisson statistics.

Droplet generating cartridge should use just once, and yes manually you can make droplets just for 8 samples at the time with droplet generator QX200/100, and the price for one package of 24 cartridges is around 200e, if you buy from Bio-Rad, also depends on package that you buy...
There is also automated droplet generator that can make droplets for 96 ddPCR reactions in 45 minutes.
