the advantages of RealTime-Glo in comparison to CellTiter-Glo are:
1. It is a non- lytic-assay
2. You can measure viability of cells in the same well several times.
3. It is ATP-independent.
I think the last point is very important. We used CellTiter-Glo for ATP measurements in macrophages and it worked very well.
Most people use it for cell viability because if a cell dies ATP levels usually drop too.
However, we observed that, while ATP levels dropped over 1, 3 and 5 hours after bacterial infection, the macrophages did not die during that time (cell death was analyzed via other techniques). That means that reduction of ATP levels (which you measure with CellTiter-Glo) does not necessarily mean that also your cells are dying (they can just use up a lot of ATP).
