Please help me in understanding the exact differences between analytical HPLC, semipreparative HPLC and preparative HPLC. I want to extract pure lutein compound from marigold oleoresin and people refer in their articles to such stuff. I'm completely illiterate to it. Please help me in understanding its functionality and how it gives pure product. How can solvent systems be fixed? Thanks.

As Aurea said, the most important difference is the purpose. With the analytical HPLC, the purpose is separation of individual components, and in preparative/semipreparative is the fractioning of groups of compounds.

• Analytical chromatography is used to determine the existence and possibly also the concentration of analyte(s) in a sample.
• Semipreparative refers to preparative LC performed on analytical (4–5 mm i.d.) or slightly larger (6–10 mm i.d.) columns. Normal injection size would be milligram- to low-gram size samples.
• Preparative chromatography is used to purify sufficient quantities of a substance for further use, rather than analysis.
