Hi there, I'm trying to detect the specific killing of target cells (a CHO-based cell line) by genetically modified T cells. The primary human T cells (both CD4 and CD8) were modified to express a target-specific receptor that leads to cellular activation upon binding. The seeded CHO cells (in a 96 well plate) express our target protein.

I have tried co-culturing different effector:target ratios (ranging from 0.02 to 10), then measuring gLuciferase or LDH in the supernatant over time, but we did not detect specific killing.

To anyone who has done this type of assay, your advice would be greatly appreciated!
