Since there should be several nucleotides between F1c and F2 (Same about B1c and B2), I doubt which would be the correct form to order primers for synthesis.
In the target sequence, F1c, F2, B1c, B2, F3 and B3 are six recognized sites which consist of the four primers and the nucleotides between F1c and F2 (B1c and B2) are usually used to design the loop primers. So, you should order the primers as FIP, BIP, F3 and B3.
Highly appreciate your kind concern to answer. You are right I have these 4 primers, but I was wondering whether I should include any sequence between F1c and F2 sequences (same about backward primers) or not. Happily, I find out that there should be a spacer sequence.
