Hello all,

I have used HEPES in the past to maintain my cells outside of CO2 incubators for extended periods of time, however some of my recent work would best be performed if the bicarbonate buffering system was not involved and simply relying on HEPES to maintain pH.

I have grown some HeLa cells in this medium (standard EMEM w 10% HIFC, Penstrep and NEAA) and noticed cells will proliferate but demonstrate a 'stretched' morphology, not the more classical appearance they usually have.

Does this imply the bicarbonate present in the media also influences cell morphology? If so, do we think that the absence of said bicarbonate would influence cell metabolism?

Any info regarding this is appreciated!
