I poured the resolving gel for a western blot without pouring the stacking gel. I left the gel with DI water on top overnight at room temperature. Is it good to use today?
Dear Imad, you can store SDS-PAGE resolving gel at 4 C without removing water layer on it. We cover gel with stretch film to prevent evaporation of water. Next day, you can pour off water and prepare stacking gel.

Hello Imad: Yes, you can use it today if the water didn't evaporate. The next time you can try using a plastic container like a small tupperware with a lid an put it in the refrigerator just to be sure.I hope it helped :)
I do store my SDS-PAGE gels overnight often, however I use two different methods for storing. I make the resolving and stacking gels with the comb completely. If I will be using the gel the next day, I will leave the gel in the casting apparatus with the comb, add a layer of Tris buffer (1 M, pH 6.8) used for the stacking gel over top to maintain the pH, and then refrigerate at 4 degrees C. If I will be using the gel after more than one day, I will remove the comb and wrap it with a wet paper towel and then plastic wrap it and place the gel at 4 degrees C.
Thus far, I have not had any problems with the resolution of the gel or how the gel runs after storage. Hope this also works for you!
