Hi Pavlina, despite the authors have used human ovarian cancer cell lines, I believe the paper below could help you out with setting up your experiments. Authors have tested the ability of increasing concentration of IFN-γ alone or in combiantion with TNF-alpha to stimulate either CXCL9 or CXCL10 in serous ovarian cancer cells. I'd suggest you starting with a similar approach by looking at which concentration works better with your cells and then move on with testing your working hypothesis. 50 ng ml-1 shows a strong to induce CXCL10 in both cell lines and I can tell you that this concentration is often used to activate a variety of immune cell types (e.g. mono/macrophages in culture).
