I am planning to examine the role of a specific protein during migration of Neural Stem Cells (NSCs). For the same I wish to either KD or KO that particular protein in NSCs. Can anyone kindly suggest whether I should use CRISPR mediated KD or shRNA mediated KO mechanism, as I am not sure when should one use CRISPR-KO and when to use shRNA-KO.
After KO or KD I will use NPCs for both in vivo expts. and in vitro transplantation studies to prove the hypothesis?

How do you want to use the shRNA mediated KD/KO? If you are going to transiently transfect cells (in utero electroporation or are you going to do it in vitro via nucleofection?) than be aware that you will lose the shRNA construct after a while.
However, maybe I did not get the question... Do you want to use a viral vector for transduction? That might be a better way and I would suggest to do this first before using CRISPR as it might be faster and more precise in the first run.
