I am currently working through a set of saliva samples for DNA extraction.

We have been following a protocol that begins by combining the sample with 60 ul of Proteinase K and 300 ul of 10% SDS, followed by an overnight incubation at 53°C. After the incubation we proceed directly into the rest of the procedure (addition of NaCl, centrifugation, isopropanol, centrifugation, ethanol, dissolve pellet in H2O, etc.).

My question is, as it is Friday, would it be alright to incubate the samples at that temperature for the duration of the weekend? Would this damage the samples? Alternatively, would it be okay to incubate the samples for approximately one hour, and then remove the samples and store at -20 for the weekend? Would this lower the DNA yield?
