Which protocol or kit do you use for plasmid isolation?

During plasmid isolation from E.coli you don't have to do any additional steps for removing endonucleases. You can use any standard kits from MN or Qiagen available for plasmid isolation which are quite easy to use.

1)    Remove enzyme  for plasmid extraction from Restriction Mix reaction

2)    Remove proteins for plasmid preparation from transformed or recombinant E Coli.

-         You can use many commercial kits to extract the plasmid of interest (eg: Qiagen kit for bacterial DNA plasmid).

-         However, if you haven’t  commercial kits you also can use Mini-prep plasmid extraction or Restriction Mix reaction followed by a phenol-chloroform extraction and ethanol precipitation and recuperate the DNA plasmid pellet in TE  pH8

-         You have to verify the plasmid purity by measuring A260/A280.

I have extraction by the Mini-prep method.

I don't use any commercial kits to extract the plasmid.
