I am currently working on a project that involves extracting cell membranes, for which we disrupt the cells using sonication. During the initial extraction process, we add protease inhibitors to prevent proteolytic degradation.
My question is regarding the subsequent step, where we intend to prepare vesicles from the isolated cell membranes. Should protease inhibitors be added again during the vesicle preparation process, or is their use only necessary during the initial extraction of the cell membranes?
