When making the SDS-PAGE running and stacking gel solution, can I save the solution without adding the TEMED and APS for future experiments? If so, how long can I save it?
Youngjoo Jo you can combine the SDS and Tris buffers and leave at room temperature as normal. However, acrylamide needs to be kept at 4 oC for long term storage. If you are in a hurry, I would set up a few tubes containing the Tris buffers and SDS which can be stored for up to a year and simply add acrylamide, APS and TEMED as you need to.
For how long? You can combine Tris, water and SDS and leave them in RT (SDS precipitates at 4°C, but if heated a little bit it becomes soluble again), and add TEMED, APS, and Acrylamide just before using them.
Yes you can save the running and stacking gel buffers at 4°C without adding the TEMED and APS for future experiment. Only thing during storage, it should not be turbid and the pH should be same.
