I need to understand the morphology of cells during cell culture .During growing cells . i found some cells rounded and not attach to substrate and other cells attach but star or rod like
First off, you need to clarify that what cells are you working with. Plant cells, for example, are attached together but animal cells are mostly adherent. About morphology of the cells, it's something you can follow with your experiment. sometimes you see different shapes at one culture but the thing is what is the shape of the majority. media component, darkness, etc can change the morphology too.
It depends on the cell type which you are using. if you see some suspended cells then either it will take time in attachment or they are dead cells. I am not sure about this because you did not mention your cell line.
I think the cells are in process of attaching, incubate for longer time. If still they fail to attain their normal morphology, then you should cross-check the growth media and type of Fetal Bovine Serum (FBS) used, and also check for any bacterial or fungal contamination.
hi ahmed just explain a bit more about your cells as in what is the cell line etc and cells are generally rounded when they are not attached to the surface so keep the cells in incubator for a longer period of time and try to read about the confluency period of your cells from ATCC and you should also check for bacterial or fungal contamination
If you are working with cardiomyocytes, or skeletal muscle cells, or even tumour cells, the round usually means, they died. Rod shape should be the proper shape for cardiomyocytes. Again, its a pity you don't specify which cells you have. All the best!
